THE APPLICATION OF BENZOTHIAZOLES IN THE DEVELOPMENT OF CHEMICAL TOOLS FOR REACTIVE SULFUR SPECIES
Reactive sulfur species (RSS) are a group of recently discovered molecules which are important to redox signal and oxidative stress related to human health and disease states. They have been found to provide protective effects in cardiovascular disease states such as hypertension, ischemia/reperfusion injury, and These species include hydrogen sulfide (H2S), hydrogen polysulfides (HSSnH), sulfur dioxide (SO2), and their effectors on proteins as post translational modifications such as persulfides (RSSH), and disulfides (RSSR), as well as sulfenic (RSOH), sulfinic (RSO2H), and sulfonic (RSO3H) acids. As such, it is vital to provide tools to better understand fundamental chemical biology of RSS and how that effects mammalian physiology. This thesis will describe the role of the unique heterocycle benzothiazole has played in providing tools to address these critical issues. First, the role of benzothiazole 2-sulfinate (BTS) as a slow releasing SO2 donor. Second, how BTS can act as a sulfinic acid transfer reagents. Finally, how the generation of a protocol to selectively sulfinylate cysteine proteins in as a generic method to study the post translation modification’s effect on protein function.
